isochron scepter and sphere of resistance

i remember hearing that to use isochron scepter with a trinisphere in play, you have to pay a total of five (2 for scepter and 3 for trinisphere) and i understand why.  my question is:  does sphere of reistance (spells cost 1 more to play) affect isochron scepter (...play it without paying its mana cost)?  does sphere of resistance affect the spell's mana cost, or is it an additional play cost?

Scepter lets you play the copy for free, but effects like trinisphere and sphere of resistance and nether void all still apply.

and defense grid also will apply then?

You're still actually playing the copy, so all relevant effects apply. Defense grid will, but only when it's not your turn.
