If I have a Painter's Servant in play naming "black", could I discard two land cards to pay the pitch cost for Soul Spike? To simplify things, are lands "colored cards" in addition to being lands with a Servant in play?

Yes.  You can also pitch artifacts.

No, but you can remove two lands in your hand from the game to play Soul Spike.
